Male and female swimmers will compete in separate events, with time handicaps applied at the start to give everyone an equal chance of winning.
It follows up on the first ever swimming race from the new Docklands facility in July, which raised €4,000 for charity partners, the RNLI and the Tiglin Lighthouse Homeless Cafe.
The new 2km LOS Docklands Challenge will see 300 swimmers of all ages splash up the Liffey from the 3Arena up under the Samuel Beckett and Sean O’Casey Bridges, before turning back at the Talbot Memorial Bridge to finish at the Jeanie Johnston beside the watersports centre pontoon.
